Research reveals role of TEFM in maintaining equilibrium between transcription and replication

It’s included in every fitness certification exam prep: mitochondria, and for good reason.

Mitochondrial DNA (mtDNA) plays a vital role in cellular energy production, and its stability is crucial for overall cell function. However, disruptions in the balance between mtDNA transcription and replication can lead to various diseases and contribute to the aging process.

A new study led by researchers in Japan has uncovered the role of mitochondrial transcription elongation factor (TEFM) in regulating this balance, offering potential insights for therapeutic advancements. For fitness professionals, understanding mitochondrial function at a deeper level can inform training strategies aimed at optimizing energy production, recovery and overall cellular health.

Published in Communications Biology on February 8, the researchers findings provide a deeper understanding of how mtDNA transcription and replication processes interact, which could have significant implications for health and disease management.

The Importance of mtDNA Regulation

Mitochondria serve as the powerhouse of the cell, generating energy through oxidative phosphorylation, a process that relies on the proper functioning of mtDNA. Unlike nuclear DNA, mtDNA is more vulnerable to mutations and instability, making its maintenance a critical area of study. Previous research has indicated that an imbalance in mtDNA transcription and replication can lead to mitochondrial dysfunction, a hallmark of numerous diseases and aging-related decline.

To investigate how TEFM influences mtDNA maintenance, the research team utilized genome editing techniques to create human cell cultures with TEFM knocked out. The study revealed that TEFM deficiency led to a significant reduction in mtDNA copy number, a decline in 7S DNA levels, and impaired strand-asynchronous replication intermediates — key indicators of mtDNA replication efficiency.

These findings suggest that TEFM plays a crucial role in regulating mtDNA replication, particularly at the heavy-strand origin of replication.

Interestingly, the absence of TEFM also triggered an increase in transcription initiation from the light-strand promoter, as evidenced by elevated levels of mitochondrial transfer RNA (tRNA Pro). However, despite this surge in transcription, the replication intermediates remained significantly reduced, indicating that TEFM is essential for maintaining the balance between these two processes.

Another significant discovery from the study was TEFM’s interaction with DNA polymerase γ (POLG), the primary enzyme responsible for mtDNA replication. This interaction suggests that TEFM not only facilitates transcription elongation but also has a direct impact on replication processes. Understanding this relationship could be key to future therapeutic strategies aimed at mitigating mtDNA-related disorders, such as mitochondrial myopathies, Leigh syndrome, MELAS syndrome, and Leber’s hereditary optic neuropathy.

Implications and Future Research

The study’s findings mark a significant advancement in mitochondrial biology, but many questions remain unanswered. Future research will aim to pinpoint the exact locations of TEFM and POLG interactions on mtDNA and further clarify their role in the overall replication mechanism.

“Our study addresses a fundamental question in mitochondrial gene expression regulation,” said Dr. Takehiro Yasukawa, associate professor at Juntendo University. “By deepening our understanding of TEFM’s role in maintaining transcription-replication balance, we hope to pave the way for new treatments targeting mtDNA-related diseases.”

Given the increasing interest in mitochondrial health within the fitness and wellness industries, this research could have long-term implications for strategies aimed at improving cellular function, longevity and disease prevention. As the scientific community continues to uncover the complexities of mtDNA regulation, the fitness and health sectors may soon benefit from these breakthroughs in ways that extend beyond traditional medical applications.

Designed for quick deployment and easy storage, the folding reformer helps fitness facilities integrate Pilates without disrupting existing programming

For many gym owners, adding new programming like Pilates comes with a major hurdle: space.

Dedicated reformer studios require significant square footage, making it difficult for fitness facilities to justify the investment. Skelcore, a Miami-based fitness and wellness company, has introduced a folding Pilates reformer designed to solve this challenge. With a compact design, the reformer allows gyms, hotels and multi-use fitness spaces to offer Pilates classes without sacrificing floor space or disrupting existing programming.

“With 53,000 gyms in the U.S. alone, many have studios that could increase their return on investment by offering Pilates as a strength modality,” Ackermann said.

The reformer’s portability makes it a practical option for gyms and other facilities with limited space.

“These reformers can be stacked safely against a wall and deployed within seconds for Pilates classes,” said Mia Bustamante, Skelcore’s product manager for Pilates.

The folding Pilates reformer is available in lightweight aluminum and American maple wood. Skelcore has also launched a range of traditional Pilates reformers, available in aluminum, oak and Canadian maple, along with an XL version in Canadian maple.

According to Skelcore, all  reformers can be customized in color and leather finish, allowing fitness facilities to tailor the equipment to their branding or design preferences.

They are built with six German-made piano wire springs for smooth resistance and aluminum tracks to enhance glide and performance. The design includes two rows of adjustable resistance hooks, allowing for customizable tension levels. The reformers feature microfiber leather upholstery and double-looped straps, enabling a wider range of exercises. Additionally, each reformer comes with a jump box to expand workout options and a safety strap for secure storage when not in use.

Skelcore has made its Pilates reformers available in 45 countries, allowing gym operators worldwide to explore Pilates as a growing revenue stream. Pulse Fitness teams up with Paralympian Ian Marsden to push for fully inclusive gym spaces, ensuring disabled athletes have equal access

Pulse Fitness has launched the Everyday Warriors campaign in collaboration with two-time Paralympian Ian Marsden to advance inclusivity in gym spaces. The initiative aims to eliminate barriers for disabled athletes and ensure fitness facilities cater to individuals of all abilities.

The campaign underscores Pulse Fitness’ commitment to fostering equitable access to fitness through its Inclusive Range of equipment, designed with accessibility at its core. With features such as swing seats for wheelchair users, multiple adjustment handles, and high-visibility color schemes for visually impaired individuals, the line currently consists of 18 strength and cardio machines.

According to the World Health Organization (WHO), an estimated 1.3 billion people, or 16% of the global population, experience significant disability. Despite this, only a small fraction of fitness facilities worldwide provide fully accessible training equipment, limiting opportunities for disabled individuals to engage in fitness activities. Adaptive fitness solutions like Pulse Fitness’ Inclusive Range are crucial in bridging this gap and promoting equitable participation in health and wellness.

Virality can boost gym memberships, but without a plan, the momentum fades fast. Here’s how fitness pros can sustain success

Social media virality can completely transform fitness businesses overnight, but gym owners and fitness professionals are discovering that “instant” or rapid success comes with its own set of challenges. While a viral workout trend or “transformation post” can lead to spikes in memberships and class bookings, it can also strain operations, resources and mental health.

Success stories, such as super viral fitness challenges on TikTok or a gym’s unique approach to training catching fire, have fueled a 27% year-over-year increase in searches for “how to go viral.” But as some gym and studio owners have learned, viral fame can be fleeting, and without the right preparation, the benefits may be short-lived.

The Temporary Boost of Virality

A viral moment can lead to a surge in interest, but it’s not always pure magic. International insurance company Hiscox  spoke to small business owners who experienced virality to share the experience, how they handled it and what they learned.

“Our TikTok video about a BBC career initiative went viral, leading to nearly 40,000 app downloads in a month,” said Samantha Hornsby, co-founder of ERIC. However, she quickly realized that many of these downloads were short-lived and the influx of new members was not sustainable. 

“A lot of people signed up just to try the class they saw online, but retention was tough,” Hornsby said.

Similarly, dance studio owner Nana Crawford saw a 118% spike in website traffic and a 30% increase in class sign-ups after a video of her business, Jiggle Fiit, went viral. But once the online attention faded, so did the new sign-ups.

While the attention and financial gain of a viral moment can be enticing, the pressure to sustain momentum can take a toll on fitness professionals’ mental well-being.

Crawford shared how the pressure to continuously post content was impacting her mental health.

“The pressure to maintain the attention took a toll. Ironically, once I started putting less pressure on myself, my videos continued to perform well,” she said.

Professional Advice

Psychotherapist Eloise Skinner advises business owners, including fitness professionals and gym operators, to stay grounded during periods of intense growth.

“Viral moments do subside, and calmer periods are likely to return. Get as much support as you can, both professionally and personally,” she suggested.

A sudden spike in demand can quickly overwhelm a solo fitness entrepreneur, leading to logistical nightmares – and potential customer dissatisfaction. To maintain quality service, some fitness businesses have adapted by hiring temporary front-desk staff, expanding class offerings and automating scheduling systems.

To help wannabe fit influencers who find themselves in the viral spotlight, Skinner offers key strategies:

Prioritize mental health: Skinner advises people to pace themselves, keep track of key feedback, and return to important comments later. “In the midst of a viral experience, things can easily get lost or overlooked,” she said.

Hire temporary staff to relieve the burden: Business expert David Horne suggests hiring temporary staff or freelancers to manage the influx of new customers. “Expanding the team provides immediate support, allowing the core team to stay focused without risking burnout,” he explained.

Focus on retention strategies: Horne stresses the importance of keeping customers engaged after the viral moment fades. 

“Offer special deals to maintain interest and analyze your metrics to understand new customer segments,” he said. Sending personalized emails to new customers can also help sustain engagement.

While preparing for viral success, fitness businesses must also consider protection.

“Having the right insurance in place, like liability coverage and equipment protection, can help safeguard your gym,” said Nick Thornhill, Direct and Partnerships Director at Hiscox. “These policies support you if workplace accidents or third-party claims arise, allowing you to focus on running your business with confidence.”

Going viral can be a sea change moment for fitness businesses, but without a measured strategy, it can also be a nightmare. By staying prepared, maintaining mental well-being and implementing specific strategies, gym owners can turn viral moments into long-term business growth rather than fleeting fame.

Outside Interactive Inc., a provider of outdoor content, mapping, events and experiences, has acquired Inntopia, a software solutions provider for the adventure travel and hospitality industry. The deal combines Outside’s audience and digital utilities with Inntopia’s booking platform, offering an integrated travel planning experience for outdoor businesses and tourism operators.

The acquisition allows Outside to incorporate Inntopia’s booking, marketing and business intelligence solutions into its system, giving ski resorts, national parks, golf and wellness resorts, and tourism bureaus a streamlined approach to reaching adventure travelers. Inntopia’s platform is used by more than 200 destinations across North America, Europe and Australia and will now integrate with Outside’s existing audience and advertising network.

“By combining our content, utilities and community with Inntopia’s booking platform technology and research, we’re creating a one-stop shop for outdoor adventure travel,” said Robin Thurston, CEO of Outside. “We see this as an opportunity to reduce friction for travelers looking to book everything from lodging to activities in one place.”

Thurston emphasized the increasing consumer shift toward experiences over material purchases, noting that “we used to be very much a purchase economy, and now people are prioritizing experiences, particularly in wellness and adventure travel.” 

The integration of Inntopia’s capabilities aligns with this trend, supporting the rise of experiential tourism across various sectors, including fitness and wellness retreats. “Our booking platform is a perfect complement to Outside’s content, utilities and community,” said Trevor Crist, CEO of Inntopia. “We’re excited to work together to create the ultimate travel platform for outdoor adventurers.”

The platform may also enable potential collaboration with fitness studios, personal trainers and wellness coaches, expanding access to outdoor adventure experiences.

“If I’m traveling somewhere for a retreat or an active vacation, why couldn’t I also book a fitness session with a local instructor or a guided outdoor workout?” Thurston said. “We’re looking at making these experiences more interconnected than people realize today.”

Inntopia’s technology simplifies the complex logistics of booking multi-component travel experiences. “Think about how difficult it is to book a full ski vacation — accommodations, lift tickets, rentals, lessons — it’s a lot. We streamline that,” Thurston explained. “And now, we can extend that concept to wellness retreats, adventure travel, and even personalized fitness experiences.”

The integration also allows for recommended curated travel experiences based on user behavior.

“I definitely think AI curation and personalization will play a significant role,” Thurston noted. “Another important factor is the addressable market — there are people who, like me, bring their bike or skis everywhere and are willing to deal with friction to have a great experience. But for many, that friction is a deterrent. As we reduce barriers and enhance personalization, we could open up a much larger market. More people might engage in these activities and even become lifelong participants if we create a seamless experience.”

The deal expands Outside’s event services and tourism partnerships while providing Inntopia’s clients with access to a broader audience. Financial terms of the acquisition were not disclosed. BrightTower, a New York-based investment banking firm, served as the exclusive financial advisor to Inntopia during the transaction.

A new peer-reviewed study from the High Altitude Exercise Physiology Program at Western Colorado University has confirmed the physiological and health benefits of the 12-3-30 workout, a treadmill routine that gained widespread popularity on social media. 

Commissioned by the American Council on Exercise (ACE), the research provides empirical support for the workout’s impact on calorie burn, cardiovascular health and exercise adherence.

The 12-3-30 workout, which involves walking at a 12% incline on a treadmill at 3.0 miles per hour for 30 minutes, first gained traction in 2020 when social media influencers touted its effectiveness as a simple and sustainable fitness routine. The study validates these claims, showing that the workout is an accessible, efficient option.

Key Findings
The study’s results highlight several benefits of the 12-3-30 workout:

  • Participants burned an average of 220 calories per session.
  • The workout received high enjoyment ratings, with all participants agreeing that it left them feeling good — essential in maintaining exercise adherence.
  • The intensity level, measured at an average of 47% of heart rate reserve, aligns with guidelines for improving cardiorespiratory fitness and reducing the risk of chronic diseases like heart disease and diabetes.

“The 12-3-30 workout is a simple yet effective way to achieve substantial health benefits,” said Lance Dalleck, PhD, lead researcher and professor at Western Colorado University. “Our findings confirm its place as a valuable tool for individuals looking to improve their fitness, whether they’re beginners or seeking a time-efficient routine.”

The findings reinforce the role of approachable, evidence-backed fitness solutions, according to Sabrena Jo, PhD, senior director of science and education at ACE. “This study shows that the 12-3-30 workout can be a practical and enjoyable way to incorporate walking into a physically active lifestyle,” she said.

There is substantial evidence that walking is an excellent exercise choice. Case in point: Recent research suggests that aiming for 8000 to 10,000 steps per day can substantially reduce the risk of CVD, diabetes, dementia and premature death.

The study highlights the workout’s potential as an effective, science-supported program for a wide variety of clients. With accessibility and enjoyment being high on the list, the routine promotes adherence and engagement, and it’s appealing for people who may be intimidated by more complex or high-intensity routines. Long known for its immune-boosting properties, elderberry is now gaining attention for another potential health benefit: improving metabolic health and aiding weight management.

A new study led by Washington State University, published in the journal Nutrients, found that consuming 12 ounces of elderberry juice daily for one week resulted in significant changes to gut microbiota, enhanced glucose tolerance and increased fat oxidation in participants.

The clinical trial involved 18 overweight adults in a randomized, placebo-controlled design. Participants consumed either elderberry juice or a placebo with similar taste and appearance, developed by North Carolina State University’s Food Innovation Lab, while following a standardized diet.

The results revealed promising shifts in gut bacteria composition, including increased levels of beneficial microbes such as firmicutes and actinobacteria, and decreased levels of harmful bacteroidetes, suggesting improved gut health, which is essential for physical and mental well-being.

Beyond gut health, elderberry juice demonstrated notable metabolic benefits. Participants who consumed the juice experienced a 24% average reduction in blood glucose levels and a 9% decrease in insulin levels, indicating improved sugar processing. Additionally, the juice enhanced fat oxidation, helping the body break down fatty acids more effectively after carbohydrate-rich meals and during exercise.

“Elderberry is an underappreciated berry, commercially and nutritionally,” said Dr. Patrick Solverson, assistant professor in WSU’s Department of Nutrition and Exercise Physiology and the study’s lead author.

“This study contributes to a growing body of evidence that elderberry, traditionally known for its immune benefits, also offers significant metabolic advantages.”

The researchers attribute these effects to elderberry’s high concentration of anthocyanins, plant-based compounds with anti-inflammatory, anti-diabetic, and antimicrobial properties. Solverson noted that elderberry contains far more anthocyanins than most other berries, requiring just six ounces of juice to match the anthocyanin content of four cups of blackberries.

However, the study’s scope does come with limitations. The small sample size of 18 participants and the short duration of the trial—just one week—mean that further research is needed to confirm the findings and understand the long-term effects of elderberry juice consumption. Additionally, variations in individual gut microbiota responses suggest that elderberry’s benefits may not be uniform across all populations.

Despite these limitations, elderberry’s potential as a “functional food” continues to generate interest. Long popular in Europe and recently experiencing a surge in demand in the U.S. during the COVID-19 pandemic, elderberry products are positioned to remain a growing market. The research team has even filed for a provisional patent to explore elderberry’s bioactive components for use in weight management and gut health applications.

Another significant investment for L Catterton is EGYM, a platform combining hardware, software and AI to support gym operators. “EGYM is arming club owners with tools to succeed,” Magliacano said. “It integrates technology to guide members and improve financial viability.” He also highlighted EGYM’s plans for a major U.S. expansion in 2025.

Magliacano sees artificial intelligence as a game-changer for the fitness industry.

A new study reveals that sitting for more than 10.6 hours a day significantly raises heart health risks, even for fit people — prompting fitness professionals to rethink strategies for combating sedentary lifestyles

Fitness professionals may need to rethink how they guide clients toward optimal health as new research highlights the dangers of prolonged sitting. A study from the Broad Institute of MIT and Harvard has found that sitting for more than 10.6 hours daily increases the risk of heart failure and cardiovascular-related deaths — even for those meeting weekly exercise recommendations.

The findings, published in the Journal of the American College of Cardiology, challenge the notion that exercise alone can fully counteract the risks of a sedentary lifestyle. Fitness professionals now face the task of integrating solutions that address both activity levels and sedentary behavior into their programs.

The Risks of Sitting Too Long

Analyzing data from 89,530 participants with an average age of 62, researchers used fitness trackers to objectively measure sedentary time. Over an 8-year follow-up, the study revealed:

  • Sitting for 10.6 hours or more daily was linked to a 40% higher risk of heart failure and a 54% increase in cardiovascular mortality, compared to those who sat less.
  • Even those who exercised for 150 minutes or more per week faced 15% higher heart failure risks and 33% increased cardiovascular-related mortality if they spent 10.6 or more hours sitting.

Dr. Shaan Khurshid, cardiologist and lead researcher, emphasized, “Too much sitting or lying down can be harmful for heart health, even for those who are active. Future guidelines and public health efforts should stress the importance of cutting down on sedentary time.”

Solutions for Fitness Professionals

Fitness professionals and health coaches have an opportunity to bridge the gap between exercise and daily movement. The American Council on Exercise (ACE) suggests applying the F.I.T.T. principle—Frequency, Intensity, Time and Type — to help clients minimize sedentary time effectively.

1. Frequency
Encourage clients to take movement breaks every 30–60 minutes. Simple actions like standing, stretching, or short walks can disrupt long periods of inactivity.

2. Intensity
Advocate light to moderate activities during these breaks, such as walking around the room or performing desk exercises, to increase circulation and engagement.

3. Time
Recommend 5–10 minutes of movement per break, aiming for an additional 30 minutes of physical activity across the day beyond their workout routine.

4. Type
Incorporate variety to sustain interest—options like standing desks, walking meetings and guided stretching can keep clients motivated and moving.

Programs that consider both structured exercise and incidental movement are essential. Fitness professionals can help clients:

  • Track their sitting time using wearable devices or apps.
  • Set realistic goals for reducing sedentary periods.
  • Integrate functional movements into daily routines to improve overall health.

A Holistic Approach to Wellness

This research provides a call to action for fit pros to expand their guidance. Educating clients about the cumulative impact of sitting, while offering practical solutions, can help them achieve better heart health and overall well-being.

Study Highlights: Wine and Cardiovascular Health

The study analyzed 1,232 participants in the PREDIMED project, a significant epidemiological study investigating the Mediterranean diet’s impact on heart health. Researchers used tartaric acid, a biomarker unique to grapes and wine, to objectively measure wine consumption — marking a departure from traditional reliance on self-reported data, which is prone to inaccuracies.

Marathon runners meticulously plan their training, gear, and nutrition — but new research from Brown University suggests they may also need to consider air quality. A study published in Sports Medicine has found that fine particulate matter in the air can significantly impact race-day performance, even for the healthiest athletes.

Analyzing data from more than 2.5 million marathon finishers across nine major U.S. marathons between 2003 and 2019, researchers discovered that higher levels of fine particulate matter correlated with slower average finish times. Male runners experienced a 32-second delay per 1 µg/m³ increase in air pollution, while female runners saw a 25-second delay.

While these delays might seem minor, they are critical for runners striving for personal bests or professional records.

“Think of all the effort a professional runner like Eliud Kipchoge puts into breaking world records,” said study author Elvira Fleury, a former graduate student at Brown and current doctoral student at Harvard University. “Our results show that air pollution can’t be ignored by those aiming to optimize performance.”

The study utilized a sophisticated spatial-temporal model to measure air pollution levels at specific points along marathon routes. The findings revealed that even pollution levels below current health standards negatively impacted athletes.

Joseph Braun, professor of epidemiology at Brown, highlighted the broader implications: “This study shows air pollution affects not just the elderly or susceptible populations, but also the fittest and healthiest among us.”

The researchers suggest that regulating emissions from power generation, transportation and industry could improve overall health outcomes and athletic performance.
